Provigil is an analeptic, used in therapy of hypersomnic conditions that are caused by narcolepsy, obstructive sleep apnea syndrome or disorder of “sleep-wake”cycle.

 

Provigil analepticis a trade name of the CNS stimulant Modafinil, which was first synthesized at the end of the last century by the French company Lafon Laboratories.

 

Clinical studies have demonstrated that Provigil effectively combats drowsiness and has a strong stimulating effect on the nervous system.

 

Provigil is approved for use only in adults. The drug is not recommended for pregnant women, nursing mothers or teenagers. Patients with cardiovascular diseasesshould take the drug with extreme caution.

  • 100mg capsule-shaped tablets
  • 200mg capsule-shaped tablets

 

Provigil tablets have a high safety profile, but their use is contraindicated in patients with hypersensitivity to Modafinil. Because of itsability to raise blood pressure, Provigil is also not recommended for patients with cardiac arrhythmia or hypertension.

 

The exact actionmechanism of Provigil is not fully understood. It is consideredthat the therapeutic effect of this analeptic is similar to the action of centrally acting sympathomimetic agents, such as Methylphenidate and Amphetamine.

 

However, the pharmacological profile of Provigil was proven to differ from sympathomimetics. It is assumed that this analeptic inhibits dopaminereuptake, thus increasing the level of this neurotransmitter in certain brain regions.

 

Some experts believe that the action of Provigil is also associated with the activation of histamine, which acts as an agonist the CNS neurotransmitters.

 

  • The recommended dose of Provigil is 200 mg per day, as a single dose. This analeptic is used in the morning or an hour before the beginning of a work shift, regardless of the meal. At the same time, studies have shown that food may slow the absorption rate of Provigil by approximately 1 hour.

 

  • It was found that the daily single Provigildose of 400mg is well-tolerated and does not cause much discomfort. However, there is no evidence that higher doses of Provigil promote an increase of the psychostimulant effect.

 

Patients with hepatic impairment should take Provigil with caution, since this analeptic may affect liver enzymes. Provigil dose for patients with severe hepatic impairment should not exceed 100mg per day.

 

Clinical studies have shown that Provigil has a stimulating effect over the CNS and mental activity. This analeptic has many advantages, which include:

But the main advantage of Provigil is its pronounced awareness-inducing effect that allows going without sleep for a long time and still feel quite comfortable.

 

It should not go unnoticed that Provigil has a low potential for abuse. Studies have shown that this analeptic is not addictive in people that use it for the first time.

 

Despite all the advantages of Provigil, the European Medicines Agency (EMA) recommends limiting the use of the drug. As the clinical use of Provigil shows, this drug may cause delayed hypersensitivity reactions.

 

Such reactions manifest most often in form of a serious rash or serious allergic reaction, feeling anxious, and depressive states.

 

Currently, Provigil is registered in more than 20 countries around the world and, in spite of its side effects, is one of the most effective drugs to help get rid of excessive sleepiness.
